Crystallization of Fructose-2,6-bisphosphatase domain of
the Human Liver Bifunctional Enzyme
The bifunctional enzyme 6-Phosphofructo-2-kinase/Fructose-2,6-bisphosphatase
is responsible for the synthesis and hydrolysis of fructose-2,6-bisphosphate,
a glycolytic stimulant. The hydrolysis of fructose-2,6-bisphosphate
by the bisphosphatase is mediated by a phospho-enzyme intermediate.
The goal of this research is to study further the change in
conformation of the human liver bisphosphatase domain (residues
250-470) of the enzyme using time-resolved crystal diffraction
as it changes through its reaction intermediates. Crystals under
the current conditions were not suitable for the completion
of the project due to the large number of protein particles
per asymmetric unit as well as the small size of the crystal.
